Albumin binding and cytotoxicity assay of nickel oxide nanoparticles against primary hippocampal neural cells

چکیده مقاله:
Nanoparticles (NPs) have been widely used in medical and therapeuticapplications. However, their albumin binding and their cytotoxicity assays havenot been well explored. In his study, the interaction of NiO NPs with human serumalbumin (HSA) was explored by circular dichroism (CD) study, molecular dockingand dynamic studies. Afterwards the cytotoxicity of NiO NPs against primaryhippocampal neural cells was explored by MTT and morphological assays. TheCD experiment revealed that NiO NPs do not stimulate any significant structuralchanges within the HSA structure. The molecular docking investigation showedthat NiO nanoclusters bind to HSA molecule through hydrophilic interactions andNiO nanoclusters with different sizes show different docking scores for interactionwith HSA. The Molecular dynamic study also revealed that minor structuralchanges in HSA structure occur after interaction with NiO NPs. Cellular assaydisplayed that incubation of NiO NPs with primary hippocampal neural cells for24 h triggered a significant cytotoxicity and morphological changes. Therefore, itmay be concluded that although NiO NPs may show a strong binding affinity toHSA and do not induce a remarkable rearrangement in the structure of HSA, theymay induce some unwanted effects on the cell viability

Cytotoxicity and apoptotic effects of nickel oxide nanoparticles in cultured HeLa cells.
The aim of this study was to observe the cytotoxicity and apoptotic effects of nickel oxide nanoparticles on human cervix epithelioid carcinoma cell line (HeLa). Nickel oxide precursors were synthesized by an nickel sulphate-excess urea reaction in boiling aqueous solution. The synthesized NiO nanoparticles (<200 nm) were investigated by X-ray diffraction analysis and transmission electron micr...
متن کاملInvestigation of cytotoxicity properties of zinc oxide nanoparticles in spherical and rod shaped on leukemia cells
In this study, we reported a method to associate doxorubicin drug on folic acid functionalized SiO2/ZnO nanoparticles (NPs) in rod and spherical shapes. The clinical usage of the anticancer drug, doxorubicin (DOX), is limited by severe side effects and cell resistance. Targeted drug delivery by binding DOX to nanoparticles could overcome these limitations. The surface functionalization of the Z...
متن کاملCytotoxic Effect of Iron Oxide Nanoparticles on Mouse Embryonic Stem Cells by MTT Assay
Background: Despite the wide range of applications, there is a serious lack of information on the impact of the nanoparticles on human health and the environment. The present study was done to determine the range of dangerous concentrations of iron oxide nanoparticle and their effects on mouse embryonic stem cells. Methods: Iron oxide nanoparticles with less than 20 nanometers diameter were en...
متن کاملThe Combined Effect of Copper Oxide and Magnesium Oxide Nanoparticles Against Water and Food Borne Bacteria
Background: The incidence of foodborne infectious diseases has been stable and even increased in many countries. Improper use of antibiotics due to the prevalence of microbial diseases has caused drug resistance. So nanotechnology has many attractive applications in the food industry, such as food preservation and food quality control. By the reason, the absorptive and antibacterial features of...

In this paper, plate-like NiO nanoparticles were prepared by one-pot solid-state thermal decomposition of nickel (II) Schiff base complex as new precursor. First, the nickel (II) Schiff base precursor was prepared by solid-state grinding using nickel (II) nitrate hexahydrate, Ni(NO3)2∙6H2O, and the Schiff base ligand N,N′-bis-(salicylidene) benzene-1,4-diamine) for 30 min without using any solv...
متن کاملExposure to titanium dioxide and other metallic oxide nanoparticles induces cytotoxicity on human neural cells and fibroblasts
The use of titanium dioxide (TiO(2)) in various industrial applications (eg, production of paper, plastics, cosmetics, and paints) has been expanding thereby increasing the occupational and other environmental exposure of these nanoparticles to humans and other species. However, the health effects of exposure to TiO(2) nanoparticles have not been systematically assessed even though recent studi...
